Survey of Buffer Solution and pH Effect on Immobilization of Lipozyme TL 100L on Celite

Abstract


In this work, we investigated that immobilization of lipase (Lipozyme TL 100 L) on celite. Celite has attractive physical properties cheap and favorable for lipase immobilization. Experimental studies are arranged by two types of buffer solutions (Tris-HCL pH range of 7 to 8.5 and phosphate pH range of 5 to 7). All immobilization processes were taken 24 h, at room temperature and 200 rpm using shaking incubator. As a result of experimental studies, at the end of 24 h, the same amount of enzyme (26 mg protein/g support) was adsorbed on celite with different immobilization yield for both of buffer solution.  The immobilization yield was 25 % for pH of 7.5 Tris-HCL buffer solutions whereas 32% for pH of 6.0 phosphate buffer solution.  According to these results we can clearly say that, Tris-HCL buffer at pH 7.5 and phosphate buffer at pH 6 is suitable for Lipozyme TL 100L lipase immobilization.
