Abstract

In this study, direct inoculation to modified semi-selective media (SMS agar-SMSA), SMS broth (SMSB) enrichment after inoculation, immunofluorescent antibody staining (IFAS), plant bioassay, enzyme-linked immunosorbent assay (ELISA) and traditional polymerase chain reaction (PCR) methods were compared. R. solanacearum was not developed on SMSA containing suspension of an artificially infected soil but a couple of soil-based bacteria were recovered. The use of modified semi-selective culture medium could be suggested to increase the availability of the low cost compared to those of other methods. However, the sensitivity of this study, although not sufficient to establish the viability of bacteria in the soil, the sensitive detection from farm soil largely depends on the sampling strategy and extraction method.
